Yes.
String contact on travel is all important, brass nuts, bone/tusq, plastic.. think or try using a brass plectrum and realise the brightness of the attack. Same thing. Why does Richie Blackmore and Malmsteen use brass nuts i wonder?
Besides, wear and tear is minimalised using quality material.

I've been thinking about making a nut from an animal bone from the woods around here. might be hard to do though. can you widdle bone? or do you just have to cut, file and sand it down?

Some people like Brass nut too , i played my old teacher tele whit brass nut and are amazing too
believe hard :
Bone : best fixed bridge guitars and acoustic guitars , tone is flesh , sappy on electrics get slight more tone and notes sound more live ... , on acoustics guitars improvise well the sound win tone and guitar win another life ...
Steel,brass : Works amazing , bright sound , flexible
Graphite : Don´t played yet
GraphTech : AMAZING at tremolos , strings own flexibility , guitar get more bright and notes get cleaner
TusQ : Don´t tried yet , i do no ...
i hate plastic shit ...

roller bridges are amazing dude , if someday i do an strato it´s will be an must
roller nut´s are cool , but .... have an problem if rust hard or lose an roller ball will need be replaced , and if someday stop be madded will be an hell to find one to replace the damaged unit

bone, vs plastic, the ongoing war.
sound wise, there is no difference,
unless you play open chords and open tunings all day long.
think about it, you fret a note, on metal, then its anchored, with another peice of metal.
thus making the bone/ high end material useless.
i did however put a brass nut on my telecaster for a few reasons.
1. it creates a similar open sound to a fretted sound instead of having the dull open plastic sound vs a brighter fretted note.
2. i use heavey strings and it should stand the test of time.
3. the strings don't catch( i put graphite in)
4. it looks baddass.
