This may be a dumb question, but...

...if I'm installing a set of hot rails in a Mustang, do I need any sort of shielding or grounding in the cavities? I'm not exactly sure if this is just for the single coils or not. Any info would be great. Thanks!

Nah, not really unless you find there is excessive noise. Unlikely if compared to the single coil.

You'll be grounding naturally when you install the pickup correctly. Easy !!

Not necessary but it doesn't hurt. Make sure your bridge (the actual bridge, not the pickup) is still properly grounded, though.

How would I do that?

Also, I have some of that shielding paint. Would it be a good idea to paint the cavities with that or is that unnecessary?

You dont need any extra grounding. Just install the pickup correctly.
